Title
PARP inhibitor and chemotherapy combination trials for the treatment of advanced malignancies: does a development pathway forward exist?

Abstract

Poly (ADP ribose) polymerase (PARP) inhibitor combinations are currently being explored as a treatment strategy for advanced cancers with some or no known DNA repair deficits in order to increase the anti-cancer potential of both the PARP inhibitor and the chemotherapy partner. However, overlapping myelosuppression necessitating lowering of the PARP inhibitor and chemotherapy doses has unknown effects on efficacy. In addition, establishment of pre-clinical efficacy will be critical in moving these combinations rationally forward for further clinical development. In this editorial, we review the mechanisms of action of PARP inhibitors and chemotherapy combinations, the clinical trials that have been performed and reported on thus far, and then discuss the challenges ahead for clinical development of these combinations.

Attention Score in Context

Attention Score in Context

This research output has an Altmetric Attention Score of 4. This is our high-level measure of the quality and quantity of online attention that it has received. This Attention Score, as well as the ranking and number of research outputs shown below, was calculated when the research output was last mentioned on 01 September 2023.
All research outputs
#9,070,438
of 30,457,780 outputs
Outputs from Annals of Oncology
#4,992
of 9,425 outputs
Outputs of similar age
#115,973
of 334,808 outputs
Outputs of similar age from Annals of Oncology
#84
of 129 outputs
Altmetric has tracked 30,457,780 research outputs across all sources so far. This one has received more attention than most of these and is in the 68th percentile.
So far Altmetric has tracked 9,425 research outputs from this source. They typically receive a lot more attention than average, with a mean Attention Score of 15.2. This one is in the 45th percentile – i.e., 45% of its peers scored the same or lower than it.
Older research outputs will score higher simply because they've had more time to accumulate mentions. To account for age we can compare this Altmetric Attention Score to the 334,808 tracked outputs that were published within six weeks on either side of this one in any source. This one has gotten more attention than average, scoring higher than 64% of its contemporaries.
We're also able to compare this research output to 129 others from the same source and published within six weeks on either side of this one. This one is in the 31st percentile – i.e., 31% of its contemporaries scored the same or lower than it.
